If you start speaking before hearing the short tone, then the recorder won't record the words spoken before it begins recording. And finally, for the PTE Retell Lecture task and any other speaking task, you shouldn't pause for longer than 3 seconds. Otherwise, the microphone is turned off for that particular question, and you cannot record anything else. For the PTE Retell lecture task, three areas of scoring are used. For the listening part, PTE does not disclose how its algorithm works, so we will focus on our speaking scoring areas, primarily content, pronunciation, and oral fluency. For Pronunciation, the PTE checks how well you can speak with a rhythm and without 'uhms' and 'aahs', and for Oral Fluency it tests how well you can speak with a rhythm and without these additional words.

Speak naturally and fluently, use correct voice modulation & pronunciation, use proper stress & speak within the specified time frame.
